Western blot analysis of extracts of various cells lines and tissues, using ACTA2 antibody CM00526HuM10-A1T at 1μg/mL. Secondary antibody: HRP Mouse Anti-Rabbit IgG (H+L) at 1：20000 dilution. Lysates/proteins: 25ug per lane.Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 3min.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human colon、human lung、monkey epididymis、monkey heart、monkey kidney、monkey liver、monkey rectum、monkey stomach、monkey tongue sections labelling ACTA2 with purified CM00526HuM10-A1T at 5μg/mL dilution. Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using citrate buffer (PH6.0). Tissue was counterstained with Hematoxylin. Mouse specific IHC polymer detection kit HRP/DAB secondary antibody was used at 1：2000 dilution. PBS instead of the primary antibody was used as the negative control.

背景信息：ACTA2 (also known as 偽-smooth muscle actin or 偽-SMA) belongs to the actin family. Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. ACTA2 is primarily expressed in vascular smooth muscle and anti-ACTA2 is commonly used to marker smooth muscle cells. This antibody may cross-react with other isoforms of actins.
